anti-CD3-MUC1 bispecific antibody

A **bispecific monoclonal antibody** targeting both the **Mucin 1 (MUC1)** antigen on tumor cells and the **CD3 antigen** on T cells, engineered to facilitate T-cell-mediated cytotoxicity against MUC1-expressing tumors.[1][3][4] This antibody bridges T cells and cancer cells, enabling the immune system—specifically cytotoxic T cells—to recognize, activate, and destroy MUC1-positive tumor cells by binding simultaneously to CD3 (on T cells) and MUC1 (on tumor cells).[1][3][4] It has been constructed using an Fab-ScFv-IgG format with an Fc-modified region to extend half-life and reduce off-target immune activation.[1] Demonstrated effects include induction of T cell activation markers (CD69, CD25, CD107a), cytokine secretion (IFN-γ, TNF-α, and IL-2), and robust in vitro and in vivo efficacy in models of breast, ovarian, pancreatic, colon, cervical, and endometrial cancers.[1] Investigated as a T-cell engager for immunotherapeutic applications across a range of epithelial malignancies, and under clinical evaluation with cytokine-induced killer (CIK) cells in liver cancer.[5][7]
