**anti-CD3 ricin A immunotoxin** is an experimental fusion toxin protein therapy comprising a monoclonal antibody directed against CD3 conjugated to the A chain of ricin, a plant-derived ribosome-inactivating protein. Upon binding to CD3 antigen on T cells, the immunotoxin is internalized, and the ricin A chain irreversibly inhibits protein synthesis within the cell, leading to apoptosis. This mechanism results in profound, though transient, depletion of T cells—especially activated T cells—and natural killer (NK) cells, as both may express CD3 or CD7 when given as part of a combination therapy. The anti-CD3 antibody also modulates the T-cell receptor complex, contributing to T cell activation-induced cell death. The drug was principally developed for steroid-refractory acute graft-versus-host disease (SR-aGVHD), where rapid and targeted immune suppression with subsequent immune reconstitution can be beneficial[1][2][3][5][6].
