Anti-CD30 CAR T-cell therapy is an adoptive cellular immunotherapy designed to target and eliminate cells expressing the CD30 (TNFRSF8) antigen. CD30 is a member of the tumor necrosis factor receptor superfamily and is a well-validated target in hematologic malignancies, particularly Hodgkin lymphoma and systemic anaplastic large cell lymphoma (ALCL). The therapy involves harvesting a patient's T cells and engineering them to express a chimeric antigen receptor (CAR) that typically includes an anti-CD30 single-chain variable fragment (scFv) linked to intracellular signaling domains such as CD3-zeta and costimulatory molecules like CD28 or 4-1BB. This specific program, investigated by The Affiliated Hospital of Xuzhou Medical University, is being evaluated in a phase 0 clinical trial (ChiCTR2100050789) for relapsed, refractory, or high-risk CD30-positive lymphoproliferative diseases, often in combination with PD-1 inhibitors to enhance persistence and efficacy by mitigating T-cell exhaustion.
