anti-CD30 NAMPTi ADC is a preclinical-stage antibody-drug conjugate (ADC) developed by Heidelberg Pharma. It consists of an anti-CD30 monoclonal antibody conjugated to an in silico-optimized nicotinamide phosphoribosyltransferase inhibitor (NAMPTi) payload via a cathepsin B-cleavable linker (such as a Val-Ala-PAB linker). By targeting CD30, which is highly expressed in certain hematological malignancies, the ADC selectively delivers the NAMPTi payload into tumor cells. Once internalized, the payload inhibits NAMPT, the rate-limiting enzyme in the NAD+ salvage biosynthesis pathway. This leads to NAD+ depletion and subsequent energy crisis, inducing cell death in both dividing and non-dividing tumor cells. Preclinical studies have demonstrated robust antitumor efficacy and prolonged survival in CD30-positive lymphoma models, including Hodgkin lymphoma.
