Anti-CD33 CAR-NK cells are a form of cell therapy in which natural killer (NK) cells are genetically engineered to express a chimeric antigen receptor (CAR) that specifically targets the CD33 antigen. CD33 is expressed on the surface of myeloid leukemia cells and some normal hematopoietic stem and progenitor cells. Upon administration, these modified NK cells bind to and selectively kill tumor cells expressing CD33. This approach aims to treat relapsed or refractory acute myeloid leukemia (AML), offering antitumor efficacy with potentially fewer side effects compared to traditional CAR-T therapies due to reduced toxicity toward normal stem/progenitor populations. The therapy can be derived from various sources such as umbilical cord blood or induced pluripotent stem cell (iPSC)-derived NK products, and may include additional modifications like 4-1BB costimulatory domains or soluble IL-15 sequences for enhanced activity[1][2][3][4][5][8].
