Overview

Anti-CD33 CART (also known as CART-33) is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy targeting CD33, a transmembrane protein expressed on the surface of myeloid lineage cells, including the majority of leukemic blasts in acute myeloid leukemia (AML). Developed by the Chinese PLA General Hospital, the therapy involves the genetic modification of a patient's own T cells to express a CAR that recognizes and binds to CD33-positive cells, leading to T-cell activation and tumor cell lysis. While showing efficacy in early-phase clinical trials (such as NCT01864902), the therapy is associated with significant myelosuppression due to the expression of CD33 on normal hematopoietic stem and progenitor cells.
