This is a third-generation chimeric antigen receptor (CAR) construct designed to target CD33-expressing cells. It incorporates an anti-CD33 single-chain variable fragment (scFv) for antigen recognition, and intracellular signaling domains from CD28, OX40, and CD3-zeta (ζ). This combination of co-stimulatory domains (CD28 and OX40) with the primary signaling domain (CD3-zeta) is intended to enhance T-cell activation, proliferation, persistence, and anti-tumor activity. It is engineered into cytokine-induced killer (CIK) cells or T cells for adoptive cell therapy, primarily investigated for acute myeloid leukemia (AML).
