anti-CD33-CLL-1 CAR-NK cells are an investigational cell therapy consisting of natural killer (NK) cells genetically engineered to express a bispecific chimeric antigen receptor (CAR) targeting both CD33 and C-type lectin-like molecule-1 (CLL-1/CLEC12A). Developed by The Affiliated Hospital of Xuzhou Medical University, this therapy is primarily aimed at treating relapsed or refractory acute myeloid leukemia (AML). CD33 is widely expressed on AML blasts, while CLL-1 is expressed on both AML blasts and leukemic stem cells (LSCs) but absent on healthy hematopoietic stem cells, making them ideal targets for dual-antigen recognition to minimize off-target effects and prevent antigen-escape-mediated relapse. The use of NK cells as a vehicle for the CAR construct may offer advantages over T-cell-based therapies, such as a lower risk of graft-versus-host disease (GvHD) and cytokine release syndrome (CRS).
