Overview

Anti-CD36 antibodies are a class of monoclonal antibodies designed to target Platelet glycoprotein 4 (CD36), a scavenger receptor that plays a key role in lipid uptake and fatty acid metabolism. In the tumor microenvironment, CD36 is selectively upregulated on regulatory T cells (Tregs), tumor-associated macrophages (TAMs), and exhausted CD8+ T cells, facilitating metabolic adaptation that drives immunosuppression. By blocking CD36-mediated lipid uptake, therapeutic antibodies such as PLT012 (developed by Pilatus Biosciences) disrupt this metabolic checkpoint, depleting immunosuppressive cell populations and restoring the effector function of cytotoxic T cells and NK cells. This approach is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors, including hepatocellular carcinoma.
