An anti-CD371 diabody is a recombinant bispecific antibody fragment designed to target CD371, also known as C-type lectin-like molecule-1 (CLL-1) or CLEC12A. CD371 is a cell surface glycoprotein predominantly expressed on myeloid lineage cells, including acute myeloid leukemia (AML) blasts and leukemic stem cells (LSCs), while being largely absent on normal hematopoietic stem cells. This expression profile makes it a highly selective target for the treatment of myeloid malignancies. The diabody architecture typically consists of two variable domains from different antibodies (e.g., anti-CD371 and anti-CD3) connected by short linkers that prevent intrachain folding, favoring the formation of a compact, bispecific molecule. In its bispecific form, the diabody functions as a T-cell engager, bridging CD371-positive tumor cells with CD3-positive T cells to trigger MHC-independent cytotoxicity and targeted cell lysis. While several anti-CD371 therapeutic candidates, including CAR-T cells and full-length bispecific antibodies, are in clinical development, the diabody format is frequently utilized in preclinical research to evaluate the efficacy of CD371-targeted T-cell redirection.
