Overview

Anti-CD38 CAR-γδ T cells is an allogeneic, off-the-shelf chimeric antigen receptor (CAR) T-cell therapy currently in preclinical development at the International Center for Cell and Gene Therapy (Fujita Health University) for the treatment of multiple myeloma. The therapy utilizes gamma-delta (γδ) T cells, which possess MHC-independent cytotoxicity, engineered via retroviral transduction to express a fully human anti-CD38 single-chain variable fragment (scFv). Unlike conventional alpha-beta (αβ) T cells, γδ T cells show minimal CD38 expression upon activation, potentially reducing the risk of fratricide. The use of a fully human CAR construct is intended to minimize immunogenicity, allowing for repeated administration. Preclinical studies have demonstrated significant in vitro and in vivo anti-tumor activity against myeloma cell lines and patient-derived samples.
