Anti-CD38 SIFbody is a preclinical therapeutic candidate developed by Momenta Pharmaceuticals (now part of Johnson & Johnson) for the treatment of multiple myeloma. It utilizes Momenta's proprietary Selective Immunomodulator of Fc receptors (SIF) platform, which leverages Fc multimerization to enhance binding to Fcγ receptors and complement. By combining the Fab region of a CD38-targeting monoclonal antibody with the SIF platform, the molecule aims to increase immune-mediated and complement-mediated cytotoxicity, including antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement-dependent cytotoxicity (CDC). In preclinical studies, it demonstrated significantly higher potency and efficacy compared to daratumumab, particularly in settings with low CD38 expression or high levels of complement inhibitors (CD55 and CD59), suggesting potential for achieving deeper minimal residual disease (MRD) negativity and overcoming resistance in multiple myeloma patients.
