Overview

anti-CD4 802-2 is a rat-derived monoclonal antibody (clone 802-2) of the IgG2b isotype that specifically targets the human CD4 coreceptor on T lymphocytes. Originally developed in the laboratory of Herman Waldmann at the University of Cambridge, this antibody is notable for its ability to induce immunological tolerance and facilitate the depletion or modulation of CD4+ T cells, rather than simply blocking the CD4-MHC class II interaction. The 802-2 clone served as the parental precursor for the humanized monoclonal antibody **TRX1**, which was developed by the biotechnology company Tolerx. TRX1 was evaluated in clinical trials for the treatment of autoimmune disorders, such as psoriasis and rheumatoid arthritis, and for the prevention of graft-versus-host disease (GvHD) and organ transplant rejection. Clinical development of the 802-2 lineage was largely halted following the dissolution of Tolerx in 2011, although the original clone remains a significant tool in preclinical immunological research.
