Anti-CD4 depletion monoclonal antibodies are targeted biologic agents designed to bind to the CD4 glycoprotein on the surface of T lymphocytes, specifically T helper cells and regulatory T cells (Tregs), to induce their rapid clearance from circulation or tissues. This depletion is typically mediated through immune effector functions such as antibody-dependent cellular cytotoxicity (ADCC) or complement-dependent cytotoxicity (CDC). In the context of immuno-oncology research, these antibodies are primarily utilized in preclinical models to eliminate immunosuppressive Tregs, thereby enhancing the anti-tumor activity of CD8+ cytotoxic T cells and improving the efficacy of other immunotherapies like PD-1/PD-L1 inhibitors. While profound CD4+ depletion is generally avoided in human clinical settings due to the risk of severe immunodeficiency, this class of antibody remains a fundamental tool in immunological research for studying T cell population dynamics and anti-tumor immunity.
