An anti-CD40 agonist antibody is a type of immunotherapeutic monoclonal antibody designed to bind and activate the CD40 receptor, a member of the tumor necrosis factor receptor (TNFR) superfamily. CD40 is widely expressed on antigen-presenting cells (APCs) such as dendritic cells, B cells, and macrophages. By mimicking the action of the natural ligand CD40L (CD154), these agonistic antibodies promote APC maturation, enhance antigen presentation, and stimulate the production of proinflammatory cytokines. This cascade leads to the activation and expansion of tumor-specific cytotoxic CD8+ T cells, effectively converting immunologically "cold" tumors into "hot" tumors. CD40 agonist antibodies are being investigated as monotherapies or in combination with immune checkpoint inhibitors, chemotherapy, or cancer vaccines for various malignancies, including pancreatic cancer, bladder cancer, melanoma, and other solid tumors.
