Anti-CD44 refers to a class of therapeutic and research agents, predominantly monoclonal antibodies, designed to target the CD44 transmembrane glycoprotein. CD44 is a cell-surface adhesion molecule and the primary receptor for hyaluronic acid (HA), playing a critical role in cell-matrix interactions, cell migration, and various signaling pathways such as JAK2/STAT3 and PI3K/Akt. It is widely recognized as a marker for cancer stem cells (CSCs) and is overexpressed in numerous malignancies, including thyroid, lung, colorectal, and gastric cancers, where it contributes to tumor progression, metastasis, and chemoresistance. Beyond oncology, anti-CD44 agents are being explored for their ability to modulate inflammatory responses in conditions like COVID-19, lupus nephritis, and osteoarthritis, as well as for their potential to block host-cell invasion by pathogens such as Plasmodium falciparum. These agents are also utilized as targeting moieties in advanced drug delivery systems, including liposomes, nanoparticles, and antibody-photoabsorber conjugates for photoimmunotherapy.
