Anti-CD44v9 antibody - monomethyl auristatin F (anti-CD44v9-MMAF) is an antibody-drug conjugate (ADC) consisting of a chimeric monoclonal antibody (clone SUM24.1, IgG1) targeting CD44 variant isoform 9 (CD44v9), conjugated to the cytotoxic payload monomethyl auristatin F (MMAF), a microtubule inhibitor. CD44v9 is a splice variant of the CD44 family that is largely absent in normal tissues but enriched in aggressive breast cancers, where it contributes to stemness, redox regulation, and therapy resistance. The drug is being investigated for the treatment of advanced breast cancer, including triple-negative, inflammatory, and ADC-resistant subtypes. It demonstrated potent, antigen-dependent cytotoxicity in vitro and dose-dependent tumor inhibition in vivo with minimal systemic toxicity. Importantly, it showed activity in cancer models resistant to existing ADCs such as trastuzumab deruxtecan, sacituzumab govitecan, and T-DM1.
