Anti-CD47 nanogel is a stimuli-responsive drug delivery system designed to provide controlled release of anti-CD47 monoclonal antibodies within the tumor microenvironment. CD47 is a transmembrane protein overexpressed on many cancer cells that functions as a "don't eat me" signal by interacting with SIRPα on macrophages to inhibit phagocytosis. While systemic CD47 blockade shows therapeutic promise, it is often limited by off-target toxicity, particularly anemia and hemagglutination due to CD47 expression on red blood cells. The nanogel formulation, typically composed of biocompatible polymers like hyaluronic acid or PEG, is engineered to release its antibody payload in response to specific triggers such as acidic pH or tumor-associated enzymes (e.g., matrix metalloproteinases). This localized delivery strategy aims to enhance the therapeutic index by concentrating the checkpoint inhibitor at the tumor site while minimizing systemic exposure and associated hematologic side effects.
