Anti-CD56 CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy developed by The Affiliated Hospital of Xuzhou Medical University. This cell therapy is engineered to target CD56 (Neural Cell Adhesion Molecule 1, NCAM1), a glycoprotein typically expressed on the surface of natural killer (NK) cells and certain T-cell subsets, which is also highly expressed in various hematologic malignancies including extranodal NK/T-cell lymphoma (ENKTL) and aggressive NK-cell leukemia (ANKL). The treatment involves the infusion of T cells modified to express a CAR that recognizes CD56, leading to the targeted destruction of CD56-positive tumor cells. It is currently being evaluated in clinical trials for patients with relapsed or refractory NK/T-cell malignancies, often following a lymphodepleting conditioning regimen of fludarabine and cyclophosphamide.
