Anti-CD6 refers to a class of monoclonal antibodies directed against the CD6 glycoprotein, a co-stimulatory receptor primarily expressed on the surface of T cells and Natural Killer (NK) cells. CD6 plays a critical role in the formation and stability of the immunological synapse and modulates lymphocyte activation, adhesion, and trafficking, primarily through its interaction with the ligand ALCAM (Activated Leukocyte Cell Adhesion Molecule, or CD166). In the context of autoimmune and inflammatory diseases, anti-CD6 antibodies (such as itolizumab) are used to inhibit the CD6-ALCAM interaction, thereby dampening the pro-inflammatory response and reducing T-cell infiltration into tissues. More recently, CD6 has emerged as a potential immune checkpoint target in oncology; research indicates that blocking CD6 can enhance the cytotoxic activity of NK cells and T cells against tumors, including multiple myeloma, by overcoming immunosuppressive signals within the tumor microenvironment.
