Overview

CD69 is a type II transmembrane glycoprotein and a member of the C-type lectin family, primarily recognized as an early activation marker on T cells, NK cells, and other lymphocytes. Beyond its role as an activation marker, CD69 functions as an immunoregulatory molecule that can limit antitumor immunity and promote T-cell exhaustion or retention within tissues. Anti-CD69 antibodies are monoclonal antibodies designed to bind and inhibit CD69, thereby reversing these inhibitory signals and enhancing the effector function of immune cells within the tumor microenvironment. Preclinical research has explored anti-CD69 therapy in various malignancies, including glioblastoma multiforme (GBM) and non-CNS cancers, often evaluating its efficacy alone or in combination with other checkpoint inhibitors like anti-PD-1 to overcome immune suppression.
