Anti-CD7 CAR-pNK cells are a form of cell therapy in which natural killer (NK) cells—often derived from allogeneic sources or the NK-92 cell line—are genetically engineered to express a chimeric antigen receptor (CAR) targeting CD7, a surface protein highly expressed on T-cell malignancies such as T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoma. The engineered NK cells are designed to recognize and kill CD7-expressing malignant cells. This approach aims to overcome challenges seen with traditional CAR-T therapies for T-cell cancers, such as fratricide and depletion of normal immune populations. Anti-CD7 CAR-pNK therapies have shown promising preclinical efficacy with reduced risk of cytokine release syndrome (CRS) and graft-versus-host disease (GvHD), due to the short lifespan and unique properties of NK cells. Anti-CD7 CAR-pNK is an investigational immunotherapy primarily being developed for relapsed/refractory T-cell malignancies, including T-cell acute lymphoblastic leukemia and lymphoma.
