Overview

**Anti-CD7 ricin A immunotoxin** is a fusion protein composed of a murine anti-CD7 monoclonal antibody chemically linked to the A-chain of the plant toxin ricin. It is designed to selectively bind to CD7-positive cells (primarily activated T cells and Natural Killer cells) and deliver ricin A, which inhibits protein synthesis and induces apoptosis. The molecule depletes CD7-expressing T cells and NK cells, suppressing immune activation and cytotoxicity. It has been primarily studied, usually in combination with an anti-CD3 ricin A immunotoxin, for the treatment of steroid-refractory acute graft-versus-host disease (SR-aGVHD) after allogeneic stem cell transplantation[1][2][3]. Development has included multi-center Phase I/II clinical trials. Side effects include manageable infusion reactions and reversible increases in creatine kinase, edema, fever, and transient cytopenias[3]. Mechanistically, it uses targeted immunosuppression by both direct cell killing (via ricin A) and functional immune inhibition (via CD7 antibody-mediated modulation).
