This anti-CD70-IL-21 immunocytokine is a novel engineered fusion protein consisting of a humanized anti-CD70 monoclonal antibody fused to interleukin-21 (IL-21). Developed by researchers at the Fox Chase Cancer Center, it is designed to target clear cell renal cell carcinoma (ccRCC), where CD70 is highly overexpressed. The IL-21 portion has been modified via site-directed mutagenesis to disrupt heparin binding, which improves target specificity and pharmacokinetic properties while reducing off-target toxicity. The molecule acts by simultaneously binding CD70 on tumor cells and activating the IL-21 receptor (IL-21R) on immune cells, thereby enhancing antibody-dependent cellular cytotoxicity (ADCC) and preventing NK cell exhaustion. Preclinical studies in humanized mouse models have demonstrated significant anti-tumor activity against CD70-positive tumors.
