Anti-CD74 monoclonal antibodies are a class of targeted biologics that bind to CD74, the invariant chain of the MHC class II complex. CD74 is highly expressed on the surface of various B-cell malignancies, such as multiple myeloma, non-Hodgkin lymphoma (NHL), and chronic lymphocytic leukemia (CLL), as well as some solid tumors like melanoma. A key characteristic of CD74 is its rapid internalization upon antibody binding, which facilitates the depletion of surface receptors and makes it an effective target for intracellular delivery of therapeutic agents. These antibodies, most notably milatuzumab (hLL1), act by blocking CD74-mediated pro-survival signaling pathways (such as the CD74/CD44 complex) and inducing apoptosis. While primarily investigated as naked antibodies to inhibit tumor growth, the anti-CD74 scaffold has also been utilized in the development of antibody-drug conjugates (ADCs) and radioimmunotherapies.
