Drug intelligence / Profile preview
Anti-CD74 neutralizing antibody is a target-specific therapeutic agent designed to bind to CD74 (HLA class II histocompatibility antigen gamma chain) and block its interaction with its cognate ligands, such as macrophage migration inhibitory factor (MIF) and D-dopachrome tautomerase (D-DT/MIF2). CD74 is a type II transmembrane glycoprotein that acts as an MHC class II chaperone and a cell surface receptor. Upon ligand binding, CD74 initiates signaling cascades (including PI3K/Akt, ERK, and NF-κB pathways) that promote cell survival, proliferation, and inflammatory cytokine secretion. By neutralizing CD74, this antibody inhibits these pro-survival and pro-metastatic pathways. In preclinical models of prostate cancer bone metastasis, targeting CD74+ macrophages with neutralizing antibodies has been shown to reduce metastasis, decrease immunosuppressive microenvironments, and increase memory B cell infiltration [1.1.0].
