Overview

Anti-CD79b CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy engineered to target CD79b, a pan B-cell surface antigen that is highly expressed across most subtypes of B-cell lymphoma. The therapy involves genetically modifying a patient's T cells, typically via lentiviral transduction, with a CAR construct derived from an anti-CD79b single-chain variable fragment (scFv) linked to co-stimulatory domains (such as 4-1BB or OX40) and the CD3ζ signaling domain. CD79b serves as an alternative or complementary target to CD19, addressing a common mechanism of resistance (CD19 antigen loss) in existing CAR T-cell therapies. Preclinical and early clinical research, primarily led by academic institutions like the University of Pennsylvania, has demonstrated potent antitumor activity in various B-cell malignancies, including diffuse large B-cell lymphoma (DLBCL) and mantle cell lymphoma (MCL).
