Anti-CD79b-RKAA-MMAE is an antibody-drug conjugate (ADC) specifically designed to target CD79b, a component of the B-cell receptor widely expressed on B-cell malignancies. This ADC consists of a monoclonal antibody targeting CD79b, engineered with an RKAA sequence modification, conjugated via a cleavable linker to the cytotoxic payload monomethyl auristatin E (MMAE), which disrupts microtubule dynamics leading to apoptosis in dividing cells. Preclinical data indicate optimal biophysical properties, a well-defined drug-to-antibody ratio of 2, and potent cytotoxic activity against B-cell lymphoma models, with an improved therapeutic index over the currently marketed anti-CD79b MMAE ADC, polatuzumab vedotin. The molecule is being developed as a safer and more efficacious treatment for diffuse large B-cell lymphoma and other B-cell non-Hodgkin lymphomas[1][4].
