Anti-CD8 refers to a class of monoclonal antibodies, antibody fragments, and imaging tracers designed to bind specifically to the CD8 coreceptor, a glycoprotein primarily expressed on the surface of cytotoxic T lymphocytes. These agents are developed for both diagnostic and therapeutic applications. In the diagnostic field, anti-CD8 immuno-PET tracers (such as 89Zr-crefmirlimab berdoxam) are used to non-invasively visualize and quantify CD8+ T-cell infiltration in tumors to predict response to immunotherapy or in the central nervous system to monitor neuroinflammatory conditions like multiple sclerosis. Therapeutically, anti-CD8 antibodies (such as the preclinical candidate PLG101) are being investigated for their ability to deplete or modulate CD8+ T cells in diseases characterized by excessive cytolytic activity, including cardiovascular conditions like myocarditis and atherosclerosis. Additionally, anti-CD8 antibodies are widely used as research reagents in preclinical models to study the role of cytotoxic T cells through targeted cell depletion.
