An anti-CD8 depleting antibody is a specialized monoclonal antibody used primarily in preclinical research to systemically eliminate CD8+ T cells in vivo. By targeting the CD8 glycoprotein on the surface of cytotoxic T lymphocytes, these antibodies induce cell death through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) or complement-dependent cytotoxicity (CDC). This systemic depletion allows researchers to investigate the specific contributions of CD8+ T cells to immune responses in various disease models, including anti-tumor immunity in glioblastoma, viral clearance, and the pathogenesis of autoimmune disorders. Common clones used in murine research include 2.43 and YTS 169.4, which are typically administered via intraperitoneal injection to achieve rapid and sustained lymphopenia of the CD8+ T cell population.
