Overview

Anti-CD9 antibodies are therapeutic monoclonal antibodies that target CD9, a member of the tetraspanin protein family. CD9 is expressed on the surface of various cancer cells, including B-acute lymphoblastic leukemia (B-ALL), gastric cancer, and several solid tumors, where its expression often correlates with poor prognosis and metastatic potential. These antibodies exert anti-tumor effects through multiple mechanisms, including the inhibition of cell proliferation, induction of apoptosis, and suppression of angiogenesis. A major hurdle in the clinical development of CD9-targeted therapies has been Fc-mediated platelet activation and aggregation, which led to thrombosis in early candidates like ALB6. Current research focuses on next-generation antibodies, such as the fully human AT1412 or Fc-engineered variants (e.g., N297A mutants), designed to maintain potent anti-leukemic and anti-tumor activity while avoiding platelet-related toxicities.
