The anti-CD90 antibody-drug conjugate (ADC) is a preclinical therapeutic candidate developed by researchers at the Fred Hutchinson Cancer Center. It utilizes a second-generation anti-CD90 single-chain variable fragment (scFv) to target CD90 (Thy-1), a surface marker expressed on hematopoietic stem cells (HSCs) and certain cancer cells. The ADC is designed for two primary applications: first, as a conditioning or selection agent to selectively deplete wild-type HSCs, thereby enriching for gene-edited HSCs that have been modified to lack the CD90 epitope; and second, as a consolidation therapy to eliminate CD90-expressing tumor cells. Preclinical studies in humanized mouse and nonhuman primate models have demonstrated its efficacy in clearing target cells without impacting the engraftment of epitope-edited, protected HSCs.
