Anti-CD90 CAR T cells are an experimental cell therapy designed to target cells expressing the CD90 (Thy-1) surface antigen. Developed by researchers at the Fred Hutchinson Cancer Center, these CAR T cells utilize a second-generation anti-CD90 single-chain variable fragment (scFv) to identify and eliminate target cells. In the context of hematopoietic stem cell (HSC) gene therapy, they are employed as a selection tool to enrich for gene-edited HSCs that have undergone CD90 ablation or binding-epitope editing. This strategy protects the modified HSCs from CAR T-mediated depletion while allowing the CAR T cells to selectively clear wild-type (non-modified) HSCs. Additionally, anti-CD90 CAR T cells are being investigated for cancer immunotherapy to target CD90-expressing tumor cells and cancer stem cells.
