Anti-CD96 monoclonal antibodies (mAbs) are a class of investigational immune checkpoint inhibitors designed to target CD96 (also known as TACTILE), a transmembrane glycoprotein expressed on T cells and natural killer (NK) cells. CD96 belongs to the immunoglobulin superfamily and interacts with its ligand, CD155 (poliovirus receptor), to deliver inhibitory signals that suppress NK cell and T cell activation and cytokine production. By blocking the CD96-CD155 interaction, anti-CD96 mAbs enhance anti-tumor immunity, promote interferon-gamma (IFN-γ) secretion, and stimulate NK and CD8+ T cell-mediated cytotoxicity. These antibodies are being evaluated, often in combination with other checkpoint inhibitors such as anti-PD-1 or anti-TIGIT therapies, for the treatment of advanced solid tumors and hematological malignancies.
