Anti-CDH3 ADC is an antibody-drug conjugate (ADC) designed to target CDH3 (P-cadherin), a cell adhesion molecule that is highly expressed in aggressive cancer types, including double-negative prostate cancer (DNPC) and triple-negative breast cancer (TNBC). Developed by researchers at the University of Notre Dame, this therapeutic agent consists of an anti-CDH3 antibody conjugated to a cytotoxic payload. In preclinical studies, the ADC demonstrated significant target-specific cytotoxicity against CDH3-high cell lines, such as DU145 and MDA-MB-468, while sparing CDH3-low or CDH3-negative cells. In vivo validation using mouse models of experimental metastasis showed that treatment with the anti-CDH3 ADC significantly reduced the metastatic burden. This program is being explored as a potential treatment strategy for metastatic prostate and breast cancers that are resistant to current standard-of-care therapies.
