Overview

Anti-CDH3 CAR-T refers to a class of chimeric antigen receptor T-cell (CAR-T) therapies engineered to target CDH3 (also known as P-cadherin), a cell adhesion protein overexpressed in various cancers. In these constructs, patient or donor T cells are genetically modified to express a synthetic receptor comprising an anti-CDH3 single-chain variable fragment (scFv) linked to intracellular co-stimulatory and signaling domains (such as 4-1BB/CD137 and CD3ζ). This enables the T cells to recognize and destroy CDH3-expressing tumor cells. Preclinical research, including studies from the University of Notre Dame presented at AACR 2025, has demonstrated significant target-specific cytotoxicity against triple-negative breast cancer (TNBC) and double-negative prostate cancer (DNPC) models.
