Overview

anti-CEA CAR-M is an investigational, allogeneic chimeric antigen receptor macrophage (CAR-M) therapy being developed by Shoreline Biosciences for the treatment of CEA-expressing solid tumors. Unlike autologous CAR-Ms derived from patient monocytes, this candidate is engineered from renewable cell sources, including induced pluripotent stem cells (iPSCs) and umbilical cord blood-derived CD34+ hematopoietic stem cells (CB-HSCs), using a standardized manufacturing platform. The cells are transduced with a lentiviral vector encoding a second-generation CAR that targets Carcinoembryonic Antigen (CEA) and incorporates unique intracellular signaling domains designed to promote and sustain a pro-inflammatory M1 phenotype. Upon binding to CEA-positive tumor cells, the CAR-Ms trigger antigen-specific phagocytosis and release pro-inflammatory cytokines, potentially overcoming the immunosuppressive microenvironment of solid tumors.
