Anti-CEA CAR-T cell therapy is an investigational autologous cell therapy where a patient's T-cells are genetically engineered to express a chimeric antigen receptor (CAR) targeting carcinoembryonic antigen (CEA), also known as CEACAM5. CEA is a cell surface glycoprotein highly expressed in various solid tumors, including colorectal, pancreatic, gastric, and lung cancers, while having limited expression in normal adult tissues. The therapy aims to direct the immune system to recognize and eliminate CEA-positive malignant cells. Clinical development, led by various institutions like Sorrento Therapeutics and Roger Williams Medical Center, has explored regional delivery methods such as hepatic artery infusion to treat liver metastases, aiming to increase local T-cell concentration and overcome the immunosuppressive tumor microenvironment of solid tumors.
