anti-CEACAM6 antibody 2A3 is a camelid-derived single-domain antibody (nanobody) that specifically recognizes the carcinoembryonic antigen-related cell adhesion molecule 6 (CEACAM6), with demonstrated cross-reactivity to CEACAM5 due to a conserved epitope within the CEACAM family.[1][3] As a 16 kDa VHH/sdAb and in an engineered heavy-chain antibody format (2A3-mFc), 2A3 has been used as a targeting moiety for molecular imaging and for chimeric antigen receptor (CAR) T-cell constructs, enabling selective recognition and killing of CEACAM6- and CEACAM5-overexpressing tumor cells in preclinical models of pancreatic and breast cancer.[1][3] Radiolabeled and fluorescently labeled 2A3 variants have shown rapid tumor uptake and favorable tumor-to-background ratios in BxPC-3 pancreatic cancer xenografts, supporting its use as a theranostic and targeting tool rather than as a stand-alone therapeutic antibody.[1][3]
