Anti-CEACAM6 HCAb-ADC is a novel antibody-drug conjugate (ADC) designed to target glycosylated Carcinoembryonic antigen-related cell adhesion molecule 6 (CEACAM6), a protein highly expressed on the surface of various epithelial-type cancers. The drug utilizes a heavy-chain antibody (HCAb) derived from a llama immune library, which provides high binding affinity (sub-nanomolar KD), enhanced thermal stability, and superior tissue penetration compared to traditional monoclonal antibodies. This HCAb is conjugated to tubulin inhibitors using hydrophilic linkers. Preclinical data presented at the AACR 2025 Annual Meeting demonstrated that the ADC effectively kills CEACAM6-positive colorectal, lung, pancreatic, and bile duct carcinoma cells at sub-nanomolar concentrations. In vivo studies showed that a single low-dose administration (0.2 mg/kg) could lead to complete tumor regression in xenograft models with minimal toxicity to normal tissues.
