Anti-claudin18.2 CAR-T cell therapy refers to a class of chimeric antigen receptor T-cell (CAR-T) therapies engineered to target Claudin 18.2 (CLDN18.2), a tight junction protein highly and selectively expressed in gastric, gastroesophageal junction (GEJ), and pancreatic adenocarcinomas. These autologous cell therapies involve isolating a patient's T cells, genetically modifying them to express a CAR construct specific to CLDN18.2, and re-infusing them to selectively eliminate CLDN18.2-positive tumor cells. The most prominent asset in this class is satricabtagene autoleucel (satri-cel, CT041), developed by CARsgen Therapeutics, which became the world's first CAR-T therapy approved for a solid tumor when China's NMPA granted approval in June 2026 for advanced CLDN18.2-positive gastric/GEJ adenocarcinoma. Other candidates, such as HEC-016, are also under early-stage clinical evaluation for various gastrointestinal malignancies.
