Anti-CLEC12A ADC refers to a class of antibody-drug conjugates (ADCs) that target C-type lectin domain family 12 member A (CLEC12A), also known as CLL-1 or CD371. CLEC12A is a transmembrane protein expressed on monocytes, acute myeloid leukemia (AML) blasts, and leukemic stem cells, but notably absent from normal hematopoietic stem cells, making it an attractive target for AML therapy. These biologics consist of a humanized anti-CLEC12A antibody conjugated to a cytotoxic payload, such as a pyrrolobenzodiazepine (PBD) dimer or a DNA-binding isoquinolidinobenzodiazepine, via a cleavable linker. Upon binding to CLEC12A-expressing cells, the ADC is internalized, and the linker is typically cleaved in the lysosome to release the payload, which then induces cell death through DNA damage. Several candidates are in development, including CLT030 and programs from ABL Bio and LegoChem Biosciences, aimed at treating AML and potentially other myeloid malignancies.
