**Anti-CLECL1 monoclonal antibody** is an investigational therapeutic antibody targeting CLECL1 (C-type lectin-like 1), a transmembrane glycoprotein expressed on a proliferative subset of chronic lymphocytic leukemia (CLL) cells, particularly the recently divided fraction (PF) marked by CXCR4^dim CD5^bright. CLECL1+ CLL cells drive tumor survival by interacting with T cells to promote Th2 differentiation and IL-4 production, fostering a supportive tumor microenvironment. These mouse-derived monoclonal antibodies, generated by immunizing with a 12-mer C-terminal CLECL1 peptide and fusing spleen cells, eradicate CLECL1+ CLL cells and block Th2 T cell and CLL growth in patient-derived xenograft (PDX) models. High CLECL1 expression correlates with shorter time to treatment and ibrutinib resistance, positioning it as a potential target for high-risk CLL.
