Overview

Anti-CLL-1 CAR-T (PD-1 knockdown) is an investigational third-generation chimeric antigen receptor (CAR) T-cell therapy. It is engineered to target C-type lectin-like molecule-1 (CLL-1, also known as CLEC12A), a protein highly expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells, while being absent on normal hematopoietic stem cells. The CAR construct contains a murine anti-CLL-1 single-chain variable fragment (scFv) and CD28 and OX40 costimulatory domains. A unique feature of this therapy is the inclusion of a short hairpin RNA (shRNA) sequence designed to knockdown the expression of programmed cell death protein 1 (PD-1). This modification aims to prevent T-cell exhaustion and enhance the CAR-T cells' ability to function within the immunosuppressive tumor microenvironment. The therapy is primarily being developed for the treatment of relapsed or refractory AML, particularly in patients who have failed prior therapies such as allogeneic stem cell transplantation or other CAR-T treatments.
