**anti-CLL1 CAR-T + CD33 CAR-T** is an investigational, dual-target chimeric antigen receptor T cell (CAR-T) therapy designed to treat acute myeloid leukemia (AML). This therapy employs engineered autologous T cells expressing CARs specific for both CLL1 (C-type lectin-like molecule-1, CLEC12A) and CD33, antigens frequently expressed on AML blasts and leukemia stem cells. The combination or tandem approach aims to enhance anti-leukemic efficacy and reduce relapse rates by targeting two independent AML antigens, thereby minimizing the risk of antigen escape. Preclinical and early clinical studies demonstrate that these dual-target CAR-T cells induce potent cytotoxicity against AML cells while sparing normal hematopoietic stem cells, though some cytotoxicity to mature myeloid cells may occur. Early clinical evidence suggests effective anti-tumor activity, with durable remissions reported in limited cases[1][6][7].
