Anti-CLL1-CD33-NKG2D bicephalic CAR-T cells is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy being developed by Xuzhou Medical University for the treatment of relapsed or refractory acute myeloid leukemia (AML). This multi-targeted therapy is designed to simultaneously recognize three distinct antigens expressed on AML cells: C-type lectin-like molecule-1 (CLL1), CD33, and ligands for the NKG2D receptor. By targeting multiple markers, the therapy aims to enhance the elimination of leukemia cells and reduce the risk of immune escape caused by antigen loss, a common challenge in single-target CAR-T therapies. The treatment involves collecting a patient's T cells via apheresis, genetically engineering them to express the specific CARs, and reinfusing them into the patient following lymphodepletion. It is currently being evaluated in Phase I/II clinical trials.
