Drug intelligence / Profile preview
anti-cMET dimeric IgA1 monoclonal antibody (cMET-dIgA) is an engineered recombinant human dimeric IgA1 monoclonal antibody designed to target the hepatocyte growth factor receptor (cMET) on the apical membrane of cyst-lining epithelial cells in polycystic kidney disease (PKD). Developed by researchers at the University of California, Santa Barbara (UCSB), this therapeutic candidate leverages the unique ability of dimeric IgA to bind the polymeric immunoglobulin receptor (pIgR)—which is highly upregulated on renal cyst-lining cells—and undergo unidirectional transcytosis into the cyst lumen. Once inside the cyst lumen, the antibody binds and antagonizes cMET, blocking hepatocyte growth factor (HGF)-mediated signaling, downregulating cell growth pathways, and inducing apoptosis specifically in cyst epithelial cells. In preclinical rodent models of PKD, cMET-dIgA treatment has demonstrated the ability to accumulate stably in cyst lumens, slow cyst growth, and improve kidney function without causing systemic or extra-renal adverse effects.
