Drug intelligence / Profile preview
anti-colchicine Anticalin (also known as Colchicalin) is a preclinical-stage engineered lipocalin biologic (Anticalin) designed as an antidote for acute colchicine poisoning. Developed using XL-protein's proprietary PASylation technology, the biologic is based on a human lipocalin 2 (Lcn2) scaffold engineered to bind and neutralize the toxic plant alkaloid colchicine in circulation with picomolar affinity (~120 pM). This binding affinity is approximately 10,000-fold stronger than colchicine's affinity for its natural target, tubulin, allowing the Anticalin to effectively scavenge and sequester the toxin in the bloodstream. To overcome the very short native plasma half-life of the Anticalin (~3.5 minutes), it is genetically fused with a 200-residue proline/alanine-rich sequence (PASylation), which extends its half-life approximately 21-fold. In preclinical rat models of acute colchicine intoxication, the PASylated Colchicalin demonstrated efficacy in accelerating symptom relief, though further dosing optimization is required to prevent rapid redistribution toxicity.
