Drug intelligence / Profile preview
An anti-CRF monoclonal antibody (such as the preclinical candidates CTRND05 or HBM9013) is an immunotherapy designed to target and neutralize corticotropin-releasing factor (CRF, also known as corticotropin-releasing hormone or CRH), the apex regulator of the hypothalamic-pituitary-adrenal (HPA) axis. By binding to CRF with picomolar affinity, the antibody blocks its interaction with CRF receptors (CRFR1 and CRFR2), thereby suppressing both basal and stress-induced activation of the HPA axis and downstream glucocorticoid (corticosterone/cortisol) release. Developed by institutions like the University of Florida and commercialized by spinouts like Andante Biologics, as well as companies like Harbour BioMed, these antibodies are being investigated for the treatment of chronic stress-related disorders, metabolic diseases, obesity, Alzheimer's disease, congenital adrenal hyperplasia (CAH), and polycystic ovarian syndrome (PCOS). In preclinical models, anti-CRF antibody treatment has demonstrated durable therapeutic effects, including selective fat mass loss, lean mass preservation, improved insulin sensitivity, and reversal of stress-induced phenotypes.
