Overview

Anti-CSF-1 refers to a class of therapeutic agents, primarily monoclonal antibodies, that target and neutralize the Colony Stimulating Factor 1 (CSF-1) ligand, also known as Macrophage Colony-Stimulating Factor (M-CSF). By binding to CSF-1, these agents prevent its interaction with the CSF-1 receptor (CSF-1R), thereby inhibiting the differentiation, proliferation, and survival of cells in the monocytic lineage, particularly immunosuppressive tumor-associated macrophages (TAMs). In oncology, anti-CSF-1 therapies are investigated for their ability to remodel the tumor microenvironment and enhance the efficacy of other treatments like radiotherapy and checkpoint inhibitors. A notable example of a specific drug in this category is lacnotuzumab (MCS110). These agents are also explored for inflammatory and proliferative conditions such as pigmented villonodular synovitis (PVNS) and tenosynovial giant cell tumor (TGCT).
